A mysterious night-time pursuit: the camera lingers on the cobblestones so typical of the historic centre of Rome. A gang of bikers is trying to kill one of the most famous pop stars in the world. We arrive in piazza della Rotonda, with the majestic Pantheon rising up in the background. The fugitive, who uncovers his face revealing himself to be Justin Bieber, is killed near the theatre of Marcellus.
Derek Zoolander (Ben Stiller), who has withdrawn from public life since losing custody of his son, is invited to Rome for a catwalk show by Alexanya Atoz (Kristen Wiig), the hottest designer of the moment. Hansel (Owen Wilson) is also invited and meets up with Derek in Rome. From above, we see St. Peter’s Square and the Vittoriano. In the car, the two models pass by piazza di Spagna and the Colosseum. At the end of the catwalk show, the pair of models are joined by Valentina Valencia (Penélope Cruz), the Interpol agent following the case of a handful of celebrities that have been killed with Zoolander’s famous look, Blue Steel, on their faces.
At Interpol’s headquarters, accessed through Valentino’s store on via Condotti, Zoolander reveals that the expression on the faces of the victims is not Blue Steel, but Aqua Vitae. With Valentina’s help, Derek finds his son at the Piccolo Anacleto orphanage, housed in the imposing Villa Aldobrandini in Frascati.
The three of them are contacted by Sting, who requests a meeting at St. Peter’s Basilica.
Sting’s information leads them to Mugatu’s prison island, which was set up in front of St. Peter’s fort on the Lazzaretto Dock in the port of Civitavecchia (the interior shots of the prison were taken at Palazzo dei Congressi).
Mugatu manages to escape through trickery and flies off by helicopter to the austere Palazzo della Civiltà Italiana, in the EUR district. The building was commissioned by Mussolini in 1937 to host great Roman works of art and is also known as the Colosseo quadrato (Square Colosseum): in the film it serves as the exterior of Alexanya Atoz’s headquarters. The interiors of the headquarters, which are spacious and intricately designed, are instead those of Palazzo dei Congressi. Here they plan to kill Derek Jr, a descendent of Steva, to drink his blood in exchange for eternal youth. Meanwhile, Derek and Valentina swim down the Tiber to arrive in time and save the boy’s life.
At the baths of Caracalla the Incrediball takes place, during which the sacrifice is set to take place in the presence of the fashion elite. While some scenes were actually shot in the underground tunnels of the ancient thermal baths, the sacrificial chamber was actually reconstructed at Cinecittà.
Other locations featured include Villa Borghese, Palazzo del Poligrafico e della Zecca dello Stato in Piazza Verdi, and the MACRO Museum of Contemporary Art.

Like its predecessor, the film centres around the life of Derek Zoolander, a once famous model who has now retreated from the public sphere. Celebrities start being killed under mysterious circumstances, and all of them had Zoolander’s famous look, Blue Steel, on their faces when they died.
